Naturforsch. 53c, 27-32 (1998); received October 9/October 31, 1997 Delayed Fluorescence, Energy Quenching, Pisum sativum , Xanthophyll Cycle Delayed chlorophyll fluorescence decay measurements on the second time scale were ap plied to investigate the state of photosystem II under different photosynthetic conditions. Leaves adapted to high and low light intensities were used to study the effects of nonphoto chemical quenching (energy quenching) on the photosynthetic state. 3-(3,4-dichlorophenyl)1,1-dimethylurea (DCMU)-treated leaves were used to characterize the photosynthetic state in the absence of a transthylakoid ApH, dithiothreitol (DTT)-treated leaves in the absence of the xanthophyll zeaxanthin. The fast decay components were the most affected by energy quenching as indicated by increased decay times. The slowest decay component was hardly affected, neither in amplitude nor in decay time. The measurements indicate a relaxation of energy quenching on the second time scale and the absence of damages in the electron transfer chain of PS II. The constant decay times of the DTT-treated leaf, comparable to those of the DCMU-treated leaf, indicate the obligatory role of zeaxanthin for most of the detected energy quenching.

In vivo target sites of nitric oxide in photosynthetic electron transport as studied by chlorophyll fluorescence in pea leaves.

The role of nitric oxide (NO) in photosynthesis is poorly understood as indicated by a number of studies in this field with often conflicting results. As various NO donors may be the primary source of discrepancies, the aim of this study was to apply a set of NO donors and its scavengers, and examine the effect of exogenous NO on photosynthetic electron transport in vivo as determined by chloro...

Inhibition of zeaxanthin formation and of rapid changes in radiationless energy dissipation by dithiothreitol in spinach leaves and chloroplasts.

Dithiothreitol, which completely inhibits the de-epoxidation of violaxanthin to zeaxanthin, was used to obtain evidence for a causal relationship between zeaxanthin and the dissipation of excess excitation energy in the photochemical apparatus in Spinicia oleracea L. In both leaves and chloroplasts, inhibition of zeaxanthin formation by dithiothreitol was accompanied by inhibition of a componen...

Topography of photosynthetic activity of leaves obtained from video images of chlorophyll fluorescence.

The distribution of photosynthetic activity over the area of a leaf and its change with time was determined (at low partial pressure of O(2)) by recording images of chlorophyll fluorescence during saturating light flashes. Simultaneously, the gas exchange was being measured. Reductions of local fluorescence intensity quantitatively displayed the extent of nonphotochemical quenching; quench coef...

A method for the rapid generation of nonsequential light-response curves of chlorophyll fluorescence.

Light-response curves (LCs) of chlorophyll fluorescence are widely used in plant physiology. Most commonly, LCs are generated sequentially, exposing the same sample to a sequence of distinct actinic light intensities. These measurements are not independent, as the response to each new light level is affected by the light exposure history experienced during previous steps of the LC, an issue par...
